基于16S rDNA的河北省鼠疫自然疫源地自毙艾鼬携带病原菌研究

摘要: 目的 对分离自河北省鼠疫自然疫源地自毙艾鼬体内的1株病原菌进行鉴定分析,为疫源地内自毙动物携带未知病原菌的研究提供参考依据。方法 利用聚合酶链式反应(PCR)对镜检形态疑似鼠疫耶尔森菌的菌落进行16S rDNA基因片段扩增并测序,测序结果经同源性比对后构建系统发育树。结果 该菌16S rDNA序列核苷酸长度为1 432 bp(登录号为OR911995)。同源性比对结果显示与序列号为NR116898的基因同源性最高(99.51%),系统发育树结果显示,与副蜂房哈夫尼亚菌处于同一进化分支,亲缘关系最近。结论 首次自河北省鼠疫自然疫源地分离到副蜂房哈夫尼亚菌。

Abstract: Objective To identify a pathogen isolated from a naturally dead Mustela eversmanni at plague foci of Hebei Province, China, so as to provide a reference for the study of unknown pathogens carried by naturally dead animals at plague foci. Methods Morphologically suspected Yersinia pestis colonies were collected for 16S rDNA gene amplification by polymerase chain reaction and sequencing. Homology alignment and phylogenetic analysis were conducted. Results The 16S rDNA sequence length was 1 432 bp (registration number: OR911995). The sequence had the highest gene homology (99.51%) with NR116898. The phylogenetic tree showed that it belonged to the same evolutionary branch as Hafnia paralvei, and they had the closest genetic relationship. Conclusion H. paralvei was for the first time isolated at natural plague foci in Hebei Province.
